I purchased some glue and have used it successfully. However, I went to use it again and the glue no longer flows when I press down. Does anyone know how to resolve this?

Glue stick applicators like this were not really intended for this purpose although the industry did not really have many choices. These are intended mainly for ink markers such as used in bingo games.
The problem is that the glue hardens arorund the sponge.
Since the glue is water soluble the best thing you can try is to snap off the sponge and soak it in warm water. Then try cleaning it with your hands under running water to see if that removes enough of the dried glue.

I tried that. But, I may have tried it too late. It did not help.
Time to get destructive 
It’s just dried glue, crunch the top with a pair of pliers, or cut off the top and use a cloth
Is the viscosity low enough to use a spray bottle, like oh an old Aqua Net bottle? Asking for a friend.
I was able to resolve this. I first tried placing the glue in a container of hot water with the top above the water. That did not work. I then found that the top section of the stick can be removed (see red in the image). Once removed, it can be cleaned out with a paper towel to restore the glue flow.
