I want to add lines to a flat surface at precise locations. Research on the internet seems that the colour paint icon is the one to use. Unfortunately, when I click on this the background grid disappears so I can only guess where to draw the line.
What is the best way of doing this?
Also, I want to look down at the top surface without any offset - is there a way of doing this without rotating the screen image?
Cmd-1 on a Mac gives you a top down view. Not sure what this on a PC.
When you say you want to add lines. Do you mean to do this in Bambu Studio for having some reference points? I would do that in whatever 3D Modelling app I am going to use. Why do you want to do that?
If you want lines on the grid I would import an SVG file that you have created in a 2D draw program and then reduce it’s thickness to 1mm. But if you are trying to establish accurate dimensions of an object it should be done in a 3D modeling or CAD app.

I am using the free version of Autodesk Fusion. I put lines on the surface using it then created an stl file for Bambu Studio but the lines were not visible.
Lines are zero width. To make lines visible you need to use two of them to define a channel then extrude or cut that channel into the model (or raise it above). To get the channel to print, it has to be at least as wide as the minimum width your printer can extrude (give or take).
You can also use the ‘thin extrude’ option in this case.
p.s. The settings in the screenshot are random and may not make sense. For example, extruding a line of 0.039 thickness to 0.4 inch tall may present problems. But hopefully you get the idea.
