A hole where, in the middle of the screw? If so, I believe they are hex/Allen wrench screws since those are used often in 3D printers. Perhaps the tool kit that came with the printer has one in the correct size. If that doesn’t help, a photo showing the part in question would help.

Don’t have 1; Seems not a hex. Will look more.

Also, how does the white line from that unit up to the intake tube stay in place inside?

The PTFE tube is held in place by an internal locking mechanism. Just push the tube in fully and check it is seated by giving a light tug on it. To unlock you push the tube in to release any tension then depress the collar and remove the tube. This is similar to water tubing John Guest fittings (not sure the generic name for them; push-fit?).
